No, they are not biological sisters. However, Rainbow Dash has taken Scootaloo "under her wing" as a surrogate sister.
Biological, no. However they do definatly have the relationship of sisters. Those two sisters are so cute together

At the end of Sleepless in Ponyville Scootaloo straight up asks Rainbow Dash to be her sister. Biologically no. Rainbow, Dash accepts. They may not be sisters by blood, but they are by heart and that's what matters most.

Obviously Dash and Scoots are not blood relatives. However, one may argue that they are essentially "sisters-in-arms".
Rainbow Dash travels with Scootaloo, protecting the younger Pony with her uncanny speed and her brilliant mind. Rainbow Dash's goal in life is to help Scotaloo find her Cutie Mark by sharpening Scootaloo's existing skills or even helping Scootaloo discover her own dormant skill, hoping that one day Scootaloo becomes the new Element of Loyalty when the time comes that Raindow Dash can defend Equestria no more. Rainbow Dash has a very calm attitude towards Scootaloo, and she doesn't rush out Scootaloo's teachings, as Scootaloo will learn what she needs to learn in due time. Despite this, Rainbow Dash prefers not to get too involved in the adventures of the Cutie Mark Crusaders, unless said adventures put Scootaloo's life at risk, at which point she will intervene to save her young pupil.

It was in quotes, so I assumed you were quoting something. If those are your own words, then all I can say is that you really need to get caught up on Season Four. There is no Element of Loyalty anymore.
1-The quotes were for dramatic effect, a way of saying "I'm gonna say something profound now". 2-I admit, I stopped watching the show after Discord was turned into stone, so I didn't know that. But if there is no more Element of Loyalty, then the Mane Six can't form the Holy Rainbow, meaning Equestria is pretty much doomed next time someone or something attacks it.
