Someone asked this before. Think about what cartoons they like already, then show them the episode that best fits that description. For example, someone who likes epic stories may want to see the first 2, while someone who likes cute cartoons may want a Fluttershy one, whereas someone who likes zany crazy comedy may like a Pinkie Pie episode.
Party of One is the one most people go by. I think Ticket Master is good for a quick introduction to the Mane Six and Applebuck Season is pretty good too, IMO. EDIT: What the ninja pirate said....

EDIT: There's a program on, on the TV, and the presenter ended the break by saying "Hello, and welcome back to 'Born to shine'", and I mistook that for "[...] welcome back to 'Fluttershy'". My addiction is taking over.
